Antibody Affinity Measurement Services

Service Overview

JWE has accumulated extensive experience in antibody and biomolecule affinity measurement. Using its proprietary Yeast Surface Display (YSD) platform, we provide high-throughput and precise affinity assessment services. Affinity is a key indicator of reversible molecular interactions and is crucial for early drug development, candidate screening, molecular interaction studies, and downstream production quality control. It is also widely applied in life sciences research and biopharmaceutical development. Leveraging a mature technology platform, JWE offers reliable and efficient one-stop solutions to meet diverse experimental needs.

Affinity Measurement Methods

JWE uses the YSD combined with FACS platform to provide fast and accurate affinity measurements. Depending on experimental requirements, biosensor technologies (BLI/SPR) and ELISA can be used to validate results, ensuring reliability. Key features include:

  • High-throughput: Screen large numbers of candidate antibodies and quickly obtain affinity distribution results
  • High sensitivity: Suitable for proteins, peptides, small molecules, and viral antigens
  • Precise quantification: With BLI/SPR, kinetic parameters such as Kd, Kon, and Koff can be obtained

Technical Overview

Method Description
YSD + FACS Display antibody fragments or VHH nanobodies on the yeast surface, and use fluorescently labeled target molecules to perform high-throughput binding and affinity clustering.
BLI/SPR Measurement Measure the binding kinetics of selected candidates, providing precise quantitative analysis for both small molecules and proteins.
ELISA Validation Supplementary validation of antibody binding activity to ensure accuracy and reduce screening costs and time.

Sample Requirements

Sample Type Requirements
Antibodies / Proteins Concentration ≥0.5 mg/mL, buffer free of Tris, EDTA, DTT
Peptides Concentration ≥1 mg/mL
Small Molecules Concentration ≥1 mg/mL, purity >90%, soluble in DMSO or water

Note: PBS, HEPPS, and other standard buffers are recommended; avoid organic solvents.
